Abstract:
Monomeric C-reactive protein (mCRP), is derived by dissociation of native CRP (nCRP) after contact with the surface of activated cells. It is a critical molecule that causes and perpetuates inflammation in association with life-threatening symptoms and disease, importantly and relevant to this project 1) causing aggregation of blood cells linked to onset of acute coronary disease and atherosclerotic plaque thrombosis; and 2) becoming stuck within tissues, within the walls/intima of major arteries, contributing significantly to worsening of unstable atherosclerotic plaque formation leading to increased chance of rupture and concomitant heart attack or stroke. CRP level in the blood correlates strongly with the above and clinical symptoms of unstable angina, heart attack, stroke and peripheral artery disease. Here, we will identify, design and create drugs by chemical synthesis and modification to satisfy this void, producing systemically deliverable pharmaceuticals blocking mCRP binding and dissociation for use in acute treatment e.g. myocardial infarction or protection of patients at high risk of thrombosis. Following thorough in vitro characterization, our proof-of-concept murine in vivo model of atherosclerosis will define their capacity for translation into clinical trials. As added value in future the same molecules could be effective in limiting inflammation and protect against tissue degeneration after stroke, traumatic brain injury and in autoimmune conditions.

Abstract:
The main objective of the project is to design a new generation of stochastic sensors based on modified graphene materials for the fast screening of biological fluids (whole blood, saliva, urine) as well as of tissue samples, in order to diagnose faster the upper gastro-intestinal cancer. In this regard, a panel of biomarkers specific to this type of cancer was selected to be analyzed using stochastic sensors. In the final stage, statistic evaluation of correlation between the panel of biomarkers found in biological fluids and tissue samples using stochastic sensors will be performed, providing the information about the possibility of diagnosis of this cancer without performing the biopsy, all only based on screening tests of biological fluids and complementary imagistic noninvasive techniques. The panel of markers proposed to be analysed is the following: - the marker that is known to be supressed during gastric and gastro-esophageal carcinogenesis - p53, it will be used as a banchmark. The other biomarkers that will be used are the following: maspin, heparanase (an endoglycosidase that is a major component of the extracellular matrix), MMP-2 and MMP-9 (two matrix metaloproteinases), HER-2 and CD26/DPP4 (a surface T cell activation antigen). Graphene materials dopped with nitrogen and boron will be synthesized and use as paste unmodified or modified with fructans, ionic liquids or aptamers for the design of stochastic sensors. The stochastic sensors will be fully characterized electrochemically, and validated against a minimum of 200 real samples from confirmed patients: biological fluids and tumoral tissues will be used for validation, but also for developing of a faster, noninvasive method of screening for fast diagnosis, avoiding in this way the biopsy.

Abstract:
Proiectul presupune dezvoltarea unui model terapeutic modern, neinvaziv, care să reducă substanțial incidența și severitatea instalării demenței în urma unui accident vascular cerebral ischemic. Pentru atingerea acestui rezultat se vor desfășura activități de cercetare experimentală realizate în laborator, pe model murin si model canin. Activitatile de cercetare vor avea loc în laboratoarele Centrului avansat de cercetări medicale și farmaceutice și în cadrul Bazei experimentale a UMF. Proiectul finanțează echipamentele specifice pentru experimentele pe model murin, echipamente pentru analize de imunohistochimie și consumabilele necesare. Proiectul urmărește și dezvoltarea de noi direcții de cercetare în cadrul UMF Tg Mureș, și anume cercetarea bolilor neurodegenerative și neuroinflamatorii, consolidarea culturii de cercetare și promovarea transferului de cunoștințe.

Abstract:
Valve reconstruction or regeneration is a daunting project for bioengineers and surgeons alike. Prosthetic heart valves and allografts are good replacement options for adult patients, but these non-living valves eventually fail after 15-20 years. Furthermore, pediatric patients cannot benefit from these choices because mechanical valves require continuous anti-coagulation, tissue valves calcify rapidly in children and allografts are in very short supply. In addition, quite often the child requires multiple interventions to replace devices which are outgrown by their rapidly growing hearts.
Tissue engineering, the science of using scaffolds, stem cells and mechanical or chemical signals may provide viable alternatives for valve replacement, reconstruction and regeneration.
Our hypothesis states that the ideal approach would combine the outstanding 3D structure provided by the aortic valve and autologous adipose derived stem cells (ADSCs) for differentiation into valvular interstitial cells (VICs) and endothelial cells (ECs) for scaffold repopulation and revitalization. The hope and expectations are that VICs would remodel and maintain matrix homeostasis while ECs would reduce valve thrombogenicity.
In our Preliminary Studies we have shown that appropriate non-antigenic scaffolds can be generated by completely removing cells from porcine aortic heart valve roots while preserving extracellular matrix integrity, that endothelial cells adhere well to acellular valve surfaces and resist bioreactor testing in vitro and that ADSCs can be coerced to differentiate into ECs and VICs thus setting the basis for generation of living valve substitutes.
We propose to validate this approach in a large animal model by collecting ADSCs from sheep, seeding acellular valve scaffolds with ECs and VICs (derived from ADSCs) and implantation as AUTOLOGOUS cell seeded scaffolds in sheep for up to 12 months to test for functionality, tissue integration, thrombogenicity and remodeling

Abstract:
Neuronal survival and protection as well as angiogenesis resulting in tissue reperfusion are key requisites for brain tissue recovery. Spatial and temporal regulation of cytoskeletal dynamics critically modulates the process of re-vascularization. Interestingly, one of the non-typical cell cycle kinases (Cdk5) has been strongly implicated in both neuroprotection and angiogenesis and is increased in stroke affected regions. Therefore modulation of Cdk5 after ischaemic stroke may have a dual action in promoting tissue survival. This project aims to produce, optimise and test a potential novel clinical therapy involving manipulation of this molecule and its cellular signalling processes.
The overall aim of the project is therefore to produce an optimized delivery system of CIP/p5 to brain tissue damaged by ischemic stroke with the intention to maximize neuronal survival and concomitantly EC survival and angiogenesis. The expected results translated into human disease would be improved reperfusion and reduced cell death and patient disability after stroke.
