Abstract:
Wound care management represents a major health care problem in the entire world and biomedical research is constantly seeking for new options with respect to promoting the healing process and to reduce the therapy cost. The goal of the project is the design, development and evaluation of new biomimetic and bioactive polymeric nanostructures (BBPNs), for the treatment of acute and chronic wounds. The innovative designed nanostructures are based on chitosan/chitosan-hyaluronic acid nanoscaffolds, which have incorporated curcumin, propolis, honey, insulin, L-arginine or allantoin, as active substances, with important biological effects such as antimicrobial, antioxidant anti-inflammatory and healing. The innovative BBPNs will be: (i) characterized using Scanning Electron Microscopy, Transmission Electron Microscopy, FT-IR spectroscopy, X-Ray Diffraction, Thermal behavior; (ii) studied in terms of physico-chemical and mechanical parameters (porosity and swelling degree, contact angle measurements, tensile strength, entrapment efficiency, rheology, water vapor transmission); (iii) kinetic studies of delivery of the active substances; (iv) in vitro assays (antimicrobial and antioxidant effects, citotoxicity and hemocompatibility, cell migration and wound closure); (v) in vivo assays (wound model induced to diabetic and nondiabetic rats). The novelty of the project is supported by the original design of the BBPNs which assure antimicrobial, anti-inflammatory and healing effects

Abstract:
Plăgile cronice reprezintă o grava problema pentru pacienţi, personalul medical cât şi pentru sistemul de sănătate. Date recente indică faptul că tratamentul plăgilor reprezintă 4% din cheltuielile sistemului de sănătate, procent care se află în continuă creştere. Cele mai comune plagi cronice sunt ulcerul diabetic, ulcerul de presiune şi ulcerul venos şi arterial al membrelor inferioare. Aceste tipuri de plăgi sunt deseori caracterizate prin hiperglicemie, inflamaţie persistentă şi nivel redus de factori de creştere şi citokine ceea ce conduce la afectarea proliferării celulare, a angiogenezei şi întărzierea epitelizării. În plus, plăgile cronice sunt deseori asociate cu infecţii bacteriene ceea ce complică cicatrizarea rănilor. În general viaţa pacienţilor ce suferă de plăgi cronice este afectată în mod semnificativ de durere, reducerea stimei de sine, infecţie şi izolare socială. În ciuda eforturilor realizate în ultimii ani, terapia actuală este departe de a asigura o vindecare adecvată şi de aceea îmbunătăţirea tratamentul plăgilor cronice a devenit o necesitate. Scopul proiectului de cercetare este dezvoltarea de noi formulări inovative pentru tratamentul local al plăgilor cronice. Aceste formulări sunt bazate pe hidrogeluri hibride de tip chitosan-acid hialuronic-bentonită, ce au incorporate o componentă activă, şi anume arginina si colagenaza, componente care sunt cunoscute pentru efecte cicatrizante. Aceste noi formulări vor fi studiate din punct de vedere al parametrilor fizico-chimici (morfologie, grad de umflare, spectroscopie FT-IR). Se va determina de asemenea efectele antimicrobiene şi antioxidante, realizîndu-se totodată studii privind cinetica eliberării in vitro a substantei active. Efectul cicatrizant va fi studiat pe model de plagă indusă la şobolanul diabetic. Este de asteptat ca aceste noi formulări inovative să rezolve atât problema infecţiei bacterine cât şi să grăbească procesul de cicatrizare, favorizând recuperarea rapidă a pacienţilor.
