Abstract:
Current project proposal is focused on the development of chitosan based materials for preparation of hydrogels and submicronic particles (micro- and/or nano-gels) for medical applications. The proposal has an interdisciplinary character which can be resumed by the following key words: thiol-ene, chitosan, cyclodextrins, hydrogels, micro/nanoparticles and drug delivery. The general objective is to prepare innovative biomaterials based chitosan and cyclodextrin through radical addition thiol-ene covalent crosslinking reaction. The experimental objectives envisaged are: finding the optimal conditions for synthesis of chemically modified chitosan and cyclodextrin with double bonds and thiol groups; preparation and characterization of novel hydrogels and micro/nanoparticles and testing the applicative potential of the new developed biomaterials. The most important elements of originality are the following: synthesis of chitosan/cyclodextrin materials with improved drug absorption/delivery kinetics; the thiol-ene coupling between chitosan and cyclodextrin derivatives with a controlled amount of functionalities available for thiol-ene reaction will be used for the first time. In addition, a systematic study of the structural factors which are influencing the covalent crosslinking will be performed.

Abstract:

Abstract:
The subject of the present proposal has a strong interdisciplinary character, being situated at the interface between chemistry, physics and biology. The main research direction will be focused on the interactions between the surface relief profile of the cells substrate and the cells response (cell fate) to the mechanical signals induced by the substrate. In the last period more and more biological studies were dedicated to this problem, especially in the case of stem cells, taking into consideration the possibility of cells differentiation using only mechanical signals. In the tissue engineering this type of interactions between the substrate and the cells will be decisive for the fabrication of smart biomaterials. Ideal candidates for new types of cells’ supports are the azo-polymers, because in their case one can change the film surface configuration, elasticity, or other mechanical parameters using only the light as external stimulus. All the other external stimuli usually used for the modification of the mechanical properties of a material (pH, temperature, salts concentration etc.) will affect the cell function making difficult to understand the cells’ response. In the case of azo-polymers, using a polarized laser source one can generate surface relief gratings on the film surface. The geometrical parameters of the gratings (amplitude and periodicity) can be very well controlled through laser operational conditions. Different chemical structure of the azo-groups will be synthesized in order to evaluate the cells adhesion capacity and the cells fate, as a function of the relief characteristics. More of than the surface relief type (grating or islands) can be changed in contact with water, creating so-called dynamic surfaces.

Abstract:
This project aims to contribute to knowledge development in the area of physicochemical characterization methods applied to oligo/peptide and/or polysaccharide systems using the modern equipment recently acquired by SANIMED International Impex SRL company and the extensive expertise of two research teams: one from the Faculty of Chemical Engineering and Environmental Protection, „Gheorghe Asachi” Technical University of Iasi and the second one from the „Petru Poni” Institute of Macromolecular Chemistry from Iasi. Highly skilled professors, researchers and specialists from both institutions will be actively involved in designing experiments, analyzing, testing and validation of SANIMED equipment and technology in order to help them adapt to the new trends by developing innovative products and processes. Special emphasis will be placed on establishing structure - properties correlations. Knowledge transfer will create premises for the development and optimization of technologies involved in the industry of cosmetics and nutritional products.
The main directions of interest are: (i) Transfer of knowledge regarding the properties of oligo / polypeptides, and polysaccharides; (ii) Studies regarding the correlation between structure of raw materials and the properties of the final products using modern analysis methods (chromatography, spectroscopy, microscopy, rheology) and SANIMED equipment; (iii) Studies on the physicochemical properties of the considered materials, using high performance equipment, present at the research units, to identify the needs of the economic agent; (iv) Studies to assess the influence of the nature of the analyzed compounds on the texture and sensory properties of the final products; (v) The use of modern methods of study in the morphological analysis of formulated products; (vi) Optimization of the formulation step and application of design of experiments; (vii) Interconnection of information in terms of chemistry, physics and informatics.
