Abstract:
Nutraceuticals (NLs) are biomolecules (carbohydrate derivatives, polyphenolics and fatty acids/structural lipids, carotenoids) that have health-promoting, disease-preventing, or medicinal properties. NLs are found in small amounts in dietary sources, thus it is necessary to develop procedures to increase their concentration and extractability. The agri-food by-products could be appropriate for using them as raw materials in the production of these industrially relevant bioactive compounds under solid-state fermentation (SSF) conditions. The aim of the present project is to develop, for the first time in the field, improved SSF processes using encapsulated fungal spores (individual and mixed-cultures), with different functions/ properties/ biosynthesis mechanisms to enrich simultaneously some abundant agro-industrial by-products (brewer's spent grain and grape pomaces) with valuable hydrophilic (polyphenolics) and lipophilic (polyunsaturated fatty acids and β-carotene) biomolecules.

Abstract:
Carotenoids are a class of lipophilic compounds with important functions in humans. The xanthophylls Lutein (Lut) and zeaxanthin (Zea), also known as macular pigments, are associated with a reduced risk of Age Related Macular Degeneration and improved cognitive function in the elderly and infants. The usual dietary ratio of Lut:Zea is approximately 5:1; a lower zeaxanthin to lutein ratio is reported as a risk factor for AMD. The health impact of carotenoids is strongly influenced by their reduced stability and by a low bioavailability, compared to other food phytochemicals. The design of carotenoid formulations compatible with food or pharmaceutical applications represents an important strategy for improvement of the stability and bioaccessibility of carotenoids. The goals of the present proposal are: (i) to determine the relationship between the type of lipid (chain lenght, unsaturation degree) and the bioaccessibility of Zeaxanthin, in model systems (pure compounds) and food systems (sea buckthorn, goji, spirulina, eggs); (ii) understanding the effect of carotenoid lipophilicity (nonesterified, esterified) on the bioaccessibility and uptake/transport of Zea using a static in vitro digestion model, intestinal cells (Caco-2/HT29-MTX co-culture) and Raman mapping; (iii) designing novel O/W emulsions/nanoemulsions (including coating with biocompatible polymers) as stable and efficient carriers of zeaxanthin for further food or nutraceutical applications.

Abstract:
The diabetic retinopathy affects the vision of people all over the world, leding to definitive blindness. In an effort to decrease the incidence of diabetic retinopathy, medical representatives and institutions are being driven to develop new strategies to improve the commonly used therapies. The current proposal is challenging and aims to push the knowledge beyond the state of the art by improving a pre-existent microsystem with attested anti-VEGF potential and transforming it into theranostic microplatforms (TPs) for multi-modal diabetic retinopathy therapy. To evaluate the biological effects of the theranostic microplatforms, two types of human cellular eye models exposed to simulated diabetic retinopathy conditions will be used. This research seeks to take use of the novel technology to develop and validate the theranostic microplatforms targeted to the VEGF protein from human retina cells and enable them to release the carried therapeutic molecules localized by using one non-invasive external irradiation source.

Abstract:
Breast cancer is a heterogeneous disease and the response to conventional therapy is closely related to hormonal status. One of the main limitations of conventional chemotherapy used for breast cancer treatment is represented by the failure or incomplete response to therapy. Thus, the identification of alternative therapies based on natural compounds aiming to reduce the toxicity exerted by conventional therapy and to prevent carcinogenesis is an urgent need.
Carotenoids are considered powerful anticancer agents that possess a number of properties such as: antioxidants, antiproliferative, proapoptotic and immune system enhancement. Numerous epidemiological studies have correlated blood carotenoid concentrations (β-cryptoxanthin, α-carotene, β-carotene, lycopene, lutein and zeaxanthin) with breast cancer risk, but the results of present studies still remain controversial. As a result, this study aims to elucidate the roles of carotenoids represented by β-cryptoxanthin, zeaxanthin and sea buckthorn lipophilic extract in the molecular mechanisms of signaling pathways in breast cancer.
Due to their extended pollen system (double conjugated bonds), carotenoids are susceptible to degradation at high temperatures, light, acid pH and in the presence of reactive oxygen species, leading to the formation of geometric isomers, epoxides or apocarotenals. Various formulations of cararotenoids have been developed in order to improve their stability, bioavalialbility or controlled release, e.g. liposomes, cyclodextrin complexes and microemulsion and continuing with newly developed nanostructures such as solid-lipid nanoparticles, nanostructured lipid carriers, nanoemulsions and nanocapsules As a result, this study aims to evaluate the efficacy of liposome encapsulated carotenoids on breast cancer cell lines (T47D, MCF-7, MDA-MB-231, BT549).

Abstract:
Coccidiosis remains one of the most economically important diseases in poultry industry. Control of coccidiosis has focused on prophylaxis with anticoccidial drugs in feed. Their extensive use has inevitably led to the development of drug resistance. The conclusion of the report in 2008 from Commision to the European Parliament was that at the present time, the use of coccidiostats as a preventive measure for the control of coccidiosis is essential, but the Commision stated that the European Commission will continue to monitor the development of new substances and techniques for the prevention. In this context through the proposed project we intend to evaluate the anticoccidial activity of an herbal product, Artemisia annua, as an effort in the development of alternative strategies of coccidiosis control and prevention. Thus, controlled battery experiments, floor pen and field trials will be done during 36 months. All the trials, except the field trial, will be done with single and mixed infections with E acervulina, E tenella and E maxima, Houghton strains.
The safety for the chicken, the activity on the immune system and microbial flora and A annua toxicity will be investigated. One of the partners will crop A annua in order to ensure the product for experiments and for field trial. The expected results of the project is to obtain a natural (herbal) product based on A annua with anti-coccidial activity (patent). Project shows promise in promoting the EU directives of reducing the chemical and pharmaceutical substances. However, its objective EU requirements are fulfilled in the field of food safety making available to poultry farmers a modern alternative for coccidiosis prophylaxis according to European legislation to coccidiostatics. The increase of Romanian researchers international visibility by article publishing and international conferences participations will be one of the desirable, collateral results.

Abstract:
The stability of carotenoids in food is affected by three major factors: light, temperature, and concentration of oxygen. Thermal treatment and light exposure induce variable losses of carotenoids, depending on their severity and length, but also on their chemical structure. They also lead to the formation of degradation products and isomers, with different colours and different biological properties. Studies pointing out the stability of xanthophyll esters (CE) are relatively scarce and the results are contradictory. The aim of this project is to evaluate the thermal and photochemical stability of carotenol esters, as well as the influence of esterification on the antioxidant activity of β-cryptoxanthin (BCR) and zeaxanthin (ZEA. Both pigments are bioactive compounds, with known beneficial effects on human health. The approach of this research is based on gradual acquisition of knowledge from the results obtained for single components (BCR and ZEA esters with different fatty acids) to the investigation of complex food matrices rich in carotenol esters (sea buckthorn and apricot. In order to accomplish this integrative approach, modern techniques will be used – LC/MS and GC/MS. The knowledge gain will improve the understanding of mechanisms of thermal and photochemical degradation of CE and could bring valuable information about the optimal conditions for storage and processing of plant derived food, in order to increase their nutritional value.
